Personally, this is the most difficult question ever. Make your choice VERRRRRRRRRRY carefully.

I think i'll give my opinions on those games(and other fighters not mentioned) first before I vote: Street Fighter started Capcom's popular fighting game series while Street Fighter II: The World Warrior(along with it's revision sequels) catapulted the series (as well as the fighting game genre) to popularity in the 1991. This was an addivtive game with lots of replay value. The characters are unique. The fighting system is excellent. And the 'less is more' method is in the special moves you pull off from each character depending on how you want to use them. Without 2-D fighting games like SFII, we wouldn't have games like Guilty Gear and KOF. ^_^ SEGA's Virtua Fighter revolutionized the fighting game genre with it's 3-D polygonal graphics engine and it became popular in it's own right. Characters were awesome. The 3-D atmosphere was unique. And the battle system is great. Without VF we wouldn't have great 3-D fighters like Tekken or Bloody Roar. ^_^ Tekken is NAMCO's answer to SEGA's Virtua Fighter. Only with interesting characters, music, the mature aspect it has, and more. ^_^ Capcom's rival SNK gave Capcom a run for it's money back in the day with their popular 2-D fighters like King Of Fighters and the Samurai Showdown series. SamSho pioneered the weapons based concept pretty good and has lead to other weapon-based fighters like Namco's Soul Edge(Soul Calibur) series. Mortal Kombat took fighting games to another level with it's graphic violence and gore. This was a controversial fighter back in the day. Peforming fatatlities after defeating an opponent was a guilty pleasure. Rarerare's Killer Instinct was a popular fighter from it's time. It borrowed concepts from Mortal Kombat(which changed the way we play fighting games with it's blood and gore) and combined it with it's unique combo system which made it stand out from other fighters. Hal lab's Super Smash Bros. was a refreshing change from traditional fighters. Not only that it's fun to play as your most favorite Nintendo characters, but the way you attack and defeat your opponent is so rewarding and different from draining an opponents life bar and defeating an opponent between rounds. Plus the 2.5D atmosphere is done well to make this fighter the best. As for my vote, each of those games you listed are good in their own right, but Street Fighter(or SFII) gets my vote because that's where it started and has taken the fighting game genre to new heights. I'm so glad Capcom made this game.

My vote goes to Super Smash Bros. While games like Street Fighter and Soul Calibur are a lot of fun, SSB is the fighting game that I've played the most.

Yeah, but the fact is, I like each of these game series listed. DarkStalkers is like Castlevania, but with Street Fighter thrown into the mix. Street Fighter and King of Fighters were awesome. Super Smash Bros. put the fighting genre on its ear. Killer Instinct Gold (For N64) began my true obsession for this genre (I played Mortal Kombats 1 and 2 first, though... They sucked). Guilty Gear had wierd but loveable characters, like the pirate girl who attacks her enemies with an anchor. Soul Calibur is what really got me interested in this whole "3D Fighter" thing. I love them all equally, but Super Smash Bros. gets one more single point simply because it has guys like Link and Fox McCloud as playable characters.

I choose SSB simply because I love the characters.

Allright, here's one: maybe I'm not a very big fan of games that have more focus on blood and guts rather than good gameplay. The characters were all lame, the story was as deep as an Oreo cookie, and the gameplay was sloggy. Of course, another reason may be this: Maybe that's just my opinion. I really don't like Mortal Kombat. I'm sorry, but that's just the way I think.

I can't really decide between Soul Calibur and SSB. SSBM is quite possibly the craziest multiplayer game ever created, but SCII is a great single player fighting game.

I'm going for Soul Calibur II. It has the best single player of a fighter, in basically all time. And multiplayer is awesome, too.
I love SSB, but the single player experience could have been better.
Tekken - The PS2's controller sucks for fighters and this game really shows it.
